Some characteristics of the immune response to sporozoites of simian and human malaria.

چکیده
The purpose of this article is to review the state of the art of immunization against malaria using the organism’s sporozoite stage, and to summarize some recent progress in this area, mainly by defining the specificity of the immune response to sporozoites of human and simian malaria. It also reports preliminary results obtained by immunizing Rhesus monkeys with irradiated sporozoites of Plasmodium cynomoZgi and attempts to correlate these findings with the results obtained in preliminary human trials (Clyde, et al., 1973). Finally, it seeks to point out a number of the gaps in our knowledge of sporozoite-induced immunit y . To summarize the state of the art on immunization with sporozoites is not a difficult task, since it has been an “unorthodox” approach followed by few investigators. In fact, most immunization attempts have used blood

Genetically attenuated, P36p-deficient malarial sporozoites induce protective immunity and apoptosis of infected liver cells.
Immunization with Plasmodium sporozoites that have been attenuated by gamma-irradiation or specific genetic modification can induce protective immunity against subsequent malaria infection. The mechanism of protection is only known for radiation-attenuated sporozoites, involving cell-mediated and humoral immune responses invoked by infected hepatocytes cells that contain long-lived, partially d...
متن کاملApoptotic Plasmodium-infected hepatocytes provide antigens to liver dendritic cells.
Malaria starts with infection of the host liver by Plasmodium sporozoites. Inoculation with radiation-attenuated Plasmodium sporozoites induces complete protection against malaria. Protection is mediated by dendritic cells (DCs) and CD8(+) T cells, but the source of parasite antigens mediating this response remains unclear. Here, we show that hepatocytes infected with irradiated Plasmodium spor...

We compare and contrast the results of immunizing mice with irradiated sporozoites of Plasmodium berghei and Plasmodium yoelii. Host genetic control of protective immunity is different in the two rodent malarias. Few mouse strains are strongly protected by P. yoelii sporozoites, while all are protected by P. berghei sporozoite immunization. The role of CD8+ T cells in the protective immune resp...
متن کاملHuman CD8+ T cells mediate protective immunity induced by a human malaria vaccine in human immune system mice
A number of studies have shown that CD8+ T cells mediate protective anti-malaria immunity in a mouse model. However, whether human CD8+ T cells play a role in protection against malaria remains unknown. We recently established human immune system (HIS) mice harboring functional human CD8+ T cells (HIS-CD8 mice) by transduction with HLA-A∗0201 and certain human cytokines using recombinant adeno-...
متن کاملThe Role of Interleukin (IL-22) in immune response to human diseases
Background and aims: IL-22 is an alpha- helical cytokine. IL-22 binds to a heterodimeric cell surface receptor composed of IL-10R2 and IL-22R1subunits. IL-22R is expressed on tissue cells, and it is absent on immune cells. L-22 and IL-10 receptor chains play a role in cellular targeting and signal transduction to selectively initiate and regulate immune responses. The aim of this study was to i...
